Enhanced Bacterial Cytological Profiling with super resolution techniques and fluorescent D-amino acids

  • Bacterial cytological profiling (BCP) is a quick one step assay to help determine antibiotic mode of action (MoA)
  • Bacteria are treated with an antibiotic and stained with fluorescent dyes associated with different cellular targets (e.g. DNA, membrane)
  • Features extracted by image analysis are processed using machine learning to associate cytological profiles with specific MoAs of known antibioitcs
  • Novel antibiotics can then be analysed and associated with known or novel pathways
  • Dyes used are FM 4-64FX, a fixable membrane stain, DAPI, a membrane permeable DNA stain, and Sytox Green, a membrane integrity indicator
